‘Load Points’ and ‘Choke Points’ as Nodes for Prioritizing Drug Targets in Pseudomonas aeruginosa

چکیده
Biological pathways information has accumulated along with Genomic sequence data. These metabolic pathways help us in understanding network robustness and complex reaction networks. They also provide a framework for improved understanding of microbial physiology and for antimicrobial drug discovery. This article is an attempt to understand the local and global properties of metabolic networks in P. aeruginosa and to identify potential drug targets through ‘load point’ and ‘choke point’ analyses. In this study, we identify 25 choke point enzymes in pathways unique to P. aeruginosa and 202 choke point enzymes in the common pathways between the pathogen and the host human. We also list top 10 choke point enzymes based on the load point values and number of shortest paths and propose them as putative targets. These data underscore the utility of systems analyses methods for understanding human metabolic network in drug discovery process and in-depth understanding of the mechanism of diseases.

Observing local and global properties of metabolic pathways: "load points" and "choke points" in the metabolic networks
MOTIVATION The local and global aspects of metabolic network analyses allow us to identify enzymes or reactions that are crucial for the survival of the organism(s), therefore directing us towards the discovery of potential drug targets. RESULTS We demonstrate a new method ('load points') to rank the enzymes/metabolites in the metabolic network and propose a model to determine and rank the bi...

متن کاملAntibiotic resistance properties and molecular characterization of Pseudomonas aeruginosa strains from patients with cystic fibrosis (CF) referred to Gholhak Pathobiology Laboratory in Tehran city during 2016-2018
Background and Objectives: Pseudomonas aeruginosa isolates from the lungs of cystic fibrosis (CF) patients are often heterogeneous and antibiotic resistant strains. Our work therefore focused on the antibiotic resistance properties of these P. aeruginosa strains isolated from Iranian patients, as well as the genetic diversity analysis by a repetitive-element-based molecular assay. Methods: This ...

The increasing trend in the occurrence of antibiotic resistance for Pseudomonas aeruginosa (a hostile bacterium growing on skin wounds especially those as a result of burn and leading to infectious conditions) has been accompanied with numerous complications and can even lead to septicemia and mortality. The spread of skin infections, especially those as a result of resistant variants of Pseudo...
متن کاملAntimicrobial Effect of Multilayered Carbon Nanotubes on Multi-Drug-Resistant Pseudomonas aeruginosa
Background: Pseudomonas aeruginosa is the primary cause of infection with impaired defense mechanisms. P. aeruginosa commonly causes nosocomial infections and is the most common pathogen isolated from patients hospitalized for longer than 1 week. We examined the antimicrobial effect of multilayered carbon nanotubes on multi-drug-resistant. Materials and methods: In this research, 20 clinical i...
